The conserved central helicase domain is shown as a black box; clusters of acidic residues are indicated by hatched boxes; and the exonuclease domain in WRN is indicated by a gray box. 学位論文要旨. 大腸菌のRecQの相同遺伝子は、出芽酵母ではSGS1のひとつしか存在しない。. ところが、ヒト細胞には大腸菌RecQと高い相同性をもつタンパク質をコードする遺伝子が5つ存在する。. それらは、我々がクローニングしたDNAヘリカーゼQ1 (RECQL1)、Bloom 出芽酵母RecQホモローグ、SGS1遺伝子破壊株がアルキル化剤に感受性である事実から本研究は出発している。またヒトに3つのRecQ遺伝子が存在し、高発癌性でしられるブルーム症候群(BLM)、早老症で知られるウエルナ-症候群(WRN)の原因遺伝子及び私が生化学的に同定したDNAヘリカーゼQ1遺伝子がある The RecQ family of DNA helicases play a key role in protecting the genome against deleterious changes.
